The widespread consumption of anorectics and combined anorectic + alcohol misuse are problems in Brazil. In order to better understand the interactive effects of ethanol (EtOH) and diethylpropion (DEP) we examined the locomotion-activating effects of these drugs given alone or in combination in mice. We also determined whether this response was affected by dopamine (DA) or opioid receptor antagonists. A total of 160 male Swiss mice weighing approximately 30 g were divided into groups of 8 animals per group. The animals were treated daily for 7 consecutive days with combined EtOH + DEP (1.2 g/kg and 5.0 mg/kg, ip), EtOH (1.2 g/kg, ip), DEP (5.0 mg/kg, ip) or the control solution coadministered with the DA antagonist haloperidol (HAL, 0.075 mg/kg, ip), the... |

Acetylsalicylic acid (ASA), the most used drug worldwide, is hydrolyzed to salicylic acid and acetate by esterases present in tissues of several species including humans. Sex differences in drug metabolism by rodent liver are documented in the literature. In this paper we report a difference in the activities of the esterases (ASA-esterase I and II) in the kidneys of male and female mice. In this species there is no difference between males and females in liver ASA-esterases (ASA-esterase I: males 38.5 ± 7.9 (N = 5) and females 31.6 ± 7.6 (N = 5) nmol of salicylic acid formed min-1 mg protein-1, P>0.05; ASA-esterase II: males 77.3 ± 17.4 (N = 5) and females 61.4 ± 15.1 (N = 5) nmol of salicylic acid formed min-1 mg protein-1, P>0.05).... |

Although the role of oxidized lipoproteins is well known in atherogenesis, the role of vitamin E supplementation is still controversial. There is also little information about cholesterol metabolism (hepatic concentration and fecal excretion) in the new models of atherosclerosis. In the present study, we evaluated the effect of moderate vitamin E supplementation on cholesterol metabolism and atherogenesis in apolipoprotein E (apo E)-deficient mice. Apo E-deficient mice were fed an atherogenic diet containing 40 or 400 mg/kg of alpha-tocopherol acetate for 6 weeks. Total cholesterol in serum and liver and 3-OH-alpha-sterols in feces, and fecal excretion of bile acids were determined and histological analyses of aortic lesion were performed. A vitamin E-rich... |

A trial was carried out to investigate the susceptibility of seven strains of mice to Strongyloides venezuelensis primary and secondary experimental infections, in order to provide the basis for genetic studies about resistance. Twelve six-week-old male inbred mice of the A/J, BALB/c, CBA/J, C3H/Hepos, C57BL/6, DBA/2 and NIH strains were infected s.c. with 2000 infective larvae. The mean worm counts (± SD) in the small intestine six days after infection were, in increasing order: 28 (± 19) in NIH; 647 (± 228) in BALB/c; 709 (± 425) in DBA/2; 731 (± 151) in C3H/Hepos, 801 (± 174) in CBA/J; 1024 (± 267) in C57BL/6 and 1313 (± 483) in A/J. C57BL/6 mice showed the highest fecal egg counts and NIH, the lowest. No eggs in fecal exams or nematodes in small... |

Previous studies have shown that rats withdrawn from long-term treatment with dopamine receptor blockers exhibit dopaminergic supersensitivity, which can be behaviorally evaluated by enhanced general activity observed in an open-field. Recently, it has been reported that co-treatment with the non-benzodiazepine anxiolytic buspirone attenuates the development of haloperidol-induced dopaminergic supersensitivity measured by open-field behavior of rats. The aims of the present study were: 1) to determine, as previously reported for rats, if mice withdrawn from long-term neuroleptic treatment would also develop dopaminergic supersensitivity using open-field behavior as an experimental paradigm, and 2) to examine if acute buspirone administration would... |

We infected NIH germ-free female mice with Helicobacter trogontum, a recently described intestinal bacterium of rats, in order to study the lesions it induced in the liver of this host. Fifteen mice were challenged with a single dose of H. trogontum (test group) and killed 6, 12 and 18 months after inoculation (5 animals/group). Nine animals were challenged with 0.85% saline alone (control group) and killed at the same times. Fragments from the liver, cecum and colon were obtained for microbiologic and histologic examination. Stool samples were also collected. H. trogontum was detected in the cecum, colon and/or stool samples of all test mice. As expected, the bacterium was not isolated from any specimen obtained from the control animals. On the other... |

Exercise training associated with robust conditioning can be useful for the study of molecular mechanisms underlying exercise-induced cardiac hypertrophy. A swimming apparatus is described to control training regimens in terms of duration, load, and frequency of exercise. Mice were submitted to 60- vs 90-min session/day, once vs twice a day, with 2 or 4% of the weight of the mouse or no workload attached to the tail, for 4 vs 6 weeks of exercise training. Blood pressure was unchanged in all groups while resting heart rate decreased in the trained groups (8-18%). Skeletal muscle citrate synthase activity, measured spectrophotometrically, increased (45-58%) only as a result of duration and frequency-controlled exercise training, indicating that endurance... |

The present work aimed to verify the effects of Baypamun®HK on the survival of albinic mice, experimentally infected with Toxoplasma gondii RH strain, treated or not with sulfadiazine-pirimetamine, as well as on the formation and the number of brain cysts. Four groups of 20 mices were inoculated with 10(5) tachizoites, subcutaneously, and undergone to different treatments. Serum tests were carried out by indirect immunofluorescence and samples of brain, liver and lung were collected from animals who died for cytological and brain cysts examinations. After 60 days, all survivors were sacrificed. Isolated Baypamun®HK didn't present a protective action against the infection by T. gondii, but its association sulfadiazine-pirimetamine, a specific treatment... |

Leishmaniasis is a disease caused by protozoa of the genus Leishmania, and visceral leishmaniasis is a form in which the inner organs are affected. Since knowledge about immunity in experimental visceral leishmaniasis is poor, we present here a review on immunity and immunosuppression in experimental visceral leishmaniasis in mouse and hamster models. We show the complexity of the mechanisms involved and differences when compared with the cutaneous form of leishmaniasis. Resistance in visceral leishmaniasis involves both CD4+ and CD8+ T cells, and interleukin (IL)-2, interferon (IFN)- gamma, and IL-12, the latter in a mechanism independent of IFN- gamma and linked to transforming growth factor (TGF)-ß production. Susceptibility involves IL-10 but not IL-4,... |

The objective of this study was to describe some changes in spermatids differentiating events during spermiogenesis, leading to spermatozoa final formation. The animals were sacrificed with ethylic acid inhalation and, then, testis fragments were collected, fixed in Karnovsky and post-fixed in osmium tetroxide. After dehydration, the sections were stained with uranyl acetate 2%. It was observed in early spermatids at Golgi phase, formation of acrosomal granule contained in the acrosomal vesicle, spheroidal nucleus with rugous reticulum around it, and Golgi apparatus made up of lamellae and vesicular structures. At cephalic hood phase, the granule suffers flattening and the marginal fossa is formed. In both phases, it was possible to observe the presence of... |

Stimulatory factors for interleukin 12 (IL-12) production associated with Babesia microti and Babesia rodhaini infected erythrocytes were examined using an in vitro assay system established, since a remarkable increase of serum IL-12 concentration and differentiation of helper T cell (Th cell) into helper T cell type 1 (Th1 cell) was observed in early phase of infection with Babesia spp. in mice. To investigate direct stimulating activity of infected erythrocytes for IL-12 production, intact splenic macrophages were co-cultured with them and examined the expression of IL-12 mRNA by reverse transcription-polymerase chain reaction (RT-PCR). Both B. microti and B. rodhaini infected erythrocytes elicited IL-12 mRNA expression in co-cultured macrophages. Since... |

The mRNA expression patterns of EGF, HB-EGF, Amphiregulin, EGF receptor, IGF-1, CSF-1, IL-1 alpha, IL-1 beta, IL-1 receptor type 1, IL-1 receptor antagonist, LIF, COX-1, COX-2, Mucin-1, calcitonin, and rat USAG-1 mouse homologue, all of which are involved in the process of conceptus implantation to the endometrium, were examined during the estrous cycle by means of real-time quantitative PCR. COX-2, HB-EGF, LIF, Mucin-1, CSF-1, IL-1 alpha, IL-1 beta, and IL-1 receptor antagonist were temporally regulated during the estrous cycle and highly expressed during the estrous stage. In the case of COX-1, EGF, IGF-1, and EGF receptor, the highest mRNA expression was during the diestrous stage. In contrast, the rat USAG-1 mouse homologue mRNA expression did not... |

Babesia spp. are major hemoprotozoan parasites in animals. Exoantigens, which are soluble antigens released by the parasites during their asexual development, have been successfully applied for the development of a vaccine against babesiosis. In this study, we examined the ability of exoantigens to induce protective immunity in mice against a rodent Babesia parasite, Babesia rodhaini. The BALB/c mice immunized with the blood plasma fraction of B. rodhaini-infected mice (B. rodhaini exoantigens) showed a significant decrease in the lethal rate when challenged with B. rodhaini. Western blot analysis with anti-B. rodhaini exoantigen immune serum specifically detected at least eleven proteins in the lysate of B. rodhaini-infected erythrocytes. Next, a cDNA... |

It is well-known that there are considerable strain differences in the relative copulation rates between male and superovulated female mice. In particular, the C57BL/6J strain of mice has a lower rate of successful copulation. We examined the effect of exposure to an electric field on sexual behavior in C57BL/6J male mice. When C57BL/6J males were exposed to a 50 Hz, 45 kV/m electric field for 30 min per day for 11 days and placed in a cage with a superovulated female of the same strain, the successful copulation rates of males was significantly improved compared with unexposed males (P<0.05). These results suggest that the exposure of C57BL/6J male mice to an electric field improves their sub-fertility activity in mating with superovulated females. |
